ปั่นแยกพลาสมาโดยวิธี Double centrifugation โดยปั่นแยกด้วยเครื่อง Centrifuge ดังนี้
  • ปั่นครั้งที่ 1 (Initial centrifugation)
          ปั่นเลือดภายใน 1 ชั่วโมงหลังเก็บ
          ความเร็วที่แนะนำ: 3,000 ×g ประมาณ 20 นาที
          แยกพลาสมาใช้ pipette พลาสติกดูดเอา ~¾ ของชั้น plasma ออก ไว้เหลือ plasma ส่วนล่างประมาณ 0.5 mL เพื่อ
          หลีกเลี่ยงการปนกับ buffy coat และ platelets
  • ปั่นครั้งที่ 2 (Double spin)
          นำ plasma ที่แยกมาใส่หลอดพลาสติก ปั่นอีกครั้งที่ 3,000 ×g 20นาที
          เพื่อลด platelet ให้เหลือน้อยกว่า 10,000/µL ซึ่งถือเป็น PPP แบ่งและแช่แข็ง (Aliquot & Freeze)
          แยกส่วน plasma แช่แข็งทันที ภายใน 4 ชั่วโมงหลังเก็บเลือด, ไม่ควรปล่อยให้ละลาย
